| Pinciple: | This mouse Periostin ELISA Kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from rat specific for Periostin has been pre-coated onto 96-well plates. Standards and test samples are added to the wells. A biotinylated detection polyclonal antibody from goat specific forPeriostin is added, followed by washing with PBS or TBS buffer. An Avidin-Biotin-Peroxidase Complex is added and the unbound conjugates are washed away with PBS or TBS buffer. HRP substrate (TMB) is used to visualize the HRP enzymatic reaction. The HRP-TMB reaction initially produces a blue color that changes yellow upon adding the acidic stop solution. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the amount of mouse Periostin within the sample that is captured by the plate. |

| Background: | Tumor necrosis factor ligand superfamily member 12 also known as TNF - related weak inducer of apoptosis (TWEAK) is a protein that in humans is encoded by the TNFSF12 gene. The protein encoded by this gene is a cytokine that belongs to the tumor necrosis factor (TNF) ligand family. This protein is a ligand for the FN14/TWEAKR receptor. This cytokine has overlapping signaling functions with TNF, but displays a much wider tissue distribution. This cytokine can induce apoptosis via multiple pathways of cell death in a cell type - specific manner. This cytokine is also found to promote proliferation and migration of endothelial cells, and thus acts as a regulator ofangiogenesis. The TNFSF12 gene lies 878 bp upstream of the putative transcriptional start site of the TNFSF13 gene on chromosome 17p13.1. |
