| Pinciple: | The Human AFGF/FGF1 (Acidic Fibroblast Growth Factor 1) ELISA Kit uses the Sandwich-ELISA method. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to human AFGF/FGF1. Standards or samples are added to the appropriate micro ELISA plate wells and combined with the specific antibody. A biotinylated detection antibody specific for human AFGF/FGF1 and Avidin-Horseradish Peroxidase (HRP) conjugate is added to each well, successively, and the plate is incubated at 37˚C. The wells are washed and the substrate solution is added. Wells containing human AFGF/FGF1, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme substrate is terminated by the addition of a sulfuric acid solution, turning the solution yellow. The optical density (OD) is measured spectrophotometrically at wavelength of 450 nm. The OD value is proportional to the concentration of the human AFGF/FGF1. You can calculate the concentration of human AFGF/FGF1 in the samples by comparing the OD of the samples to the standard curve. |
